Krypto the Super-Dog has been Superman's inseparable companion since both escaped Krypton as infants. When Superman begins a serious relationship with Lois Lane, Krypto grows jealous and anxious about being replaced. Superman takes Krypto to a Metropolis animal shelter to find him a canine companion, where they meet Ace, a tough Boxer dog repeatedly attempting escape. Meanwhile, Lex Luthor tries to retrieve a chunk of orange kryptonite from space via tractor beam, but Superman and the Justice League foil him immediately. At LexCorp, Lulu, a hairless guinea pig who has been a test subject for years, manages to grab a fragment of the orange kryptonite for herself. It grants her powerful telekinesis. As the shelter catches fire during the chaos, the four resident animals—Ace, PB (a potbellied pig), Merton (a nearsighted turtle), and Chip (a paranoid squirrel)—are inadvertently exposed to the kryptonite and gain superpowers of their own: Ace becomes super-strong and nearly invulnerable, PB can grow or shrink to any size, Merton gains super-speed, and Chip develops electrokinesis. Lulu escapes the shelter and enacts a scheme to capture Superman, which she accomplishes. Krypto is tricked into eating green kryptonite hidden in cheese, stripping him of his powers. Desperate and depowered, Krypto reluctantly teams up with the newly powered shelter animals to rescue Superman. Lulu meanwhile assembles an army of fellow guinea pigs, uses them to capture the entire Justice League, and breaks Lex Luthor out of Stryker's Island prison—only for Luthor to immediately betray her. At LexCorp the team battles Whiskers, a kitten with the ability to conjure weapons, whom they eventually trap. Krypto's powers gradually return as the green kryptonite passes through his digestive system. Lulu escalates her plan, launching the captive Justice League into space aboard a rocket and then targeting Lois Lane for assassination. The shelter pets work together to free the Justice League heroes while Krypto intercepts and saves Lois. In the final confrontation, Lulu absorbs a massive dose of kryptonite and transforms into a gigantic, kaiju-sized monster. Krypto sacrifices himself by executing a 'Solar Paw Punch' to stop her, and is saved from death by Ace's invulnerability shielding him. The defeated Lulu is trapped inside a hot dog stand by the reformed guinea pig army. In the aftermath, each shelter animal is adopted by a Justice League member, and the group officially becomes the 'League of Super-Pets.' Post-credits scenes show Luthor imprisoned again and Krypto crossing paths with Black Adam.
